Biologische activiteit

Beschrijving Chlorpromazine (CPZ) is een typisch antipsychoticum met lage potentie dat wordt gebruikt om psychotische stoornissen zoals schizofrenie te behandelen. Deze verbinding remt in vitro de 5-HT(2A) receptor en dopamine D2. Het remt ook het sodium channel en blokkeert HERG potassium channels met een IC50 van 21,6 μM.
In vitro

Chlorpromazine veroorzaakt een lichte afname van de sEV-opname in slechts één MM-cellijn, wat aangeeft dat de opname van BMSC-afgeleide sEV's door suspensie-MM-cellen mogelijk niet afhankelijk is van deze op de verbinding gerichte receptoren.
In Vivo

Chlorpromazine onderdrukt de groei van endometriumkanker (EC)-tumoren in vivo bij een lage dosis zonder tekenen van toxiciteit.

  • Methode

    MM1S cells pre-treated with heparin, cytochalasin D (Cyto D), dynasore, genistein, or chlorpromazine (CPZ) at the indicated concentrations for 30 min are cultured with 50 μg/mL DID-labeled BMSC-derived sEVs for another 4 h. The mean fluorescence intensity of DID in these cells is determined using flow cytometry. RPMI 8226, U266, or H929 cells are treated with heparin, Cyto D, dynasore, genistein, or this compound at the indicated concentration for 4.5 h in the absence of sEVs and their cell viability is measured.
