Beschrijving IOX1 is een potente en breedspectrumremmer van 2OG oxygenasen, inclusief de JmjC demethylasen. Deze verbinding is een remmer van ALKBH5.

In vitro

IOX1 verhoogt H3K9me3-niveaus in HeLa-cellen via KDM4A-remming zonder significant effect op de cellevensvatbaarheid. Deze verbinding vertoont een lagere werkzaamheid in HeLa-cellen als gevolg van een lage celpermeabiliteit, terwijl het n-octylesterderivaat de celpermeabiliteit verbetert.

  • AlphaScreen Assay

    Alle reagentia worden verdund in 50 mM HEPES, 0,1% BSA, pH 7,5 aangevuld met 0,01% Tween20 en tot kamertemperatuur laten equilibreren voordat ze aan de platen worden toegevoegd. Katalytische omzettingstesten worden uitgevoerd in volumes van 10 μL in 384-wells platen met laag volume bij kamertemperatuur. De reactie bestond uit enzym (5 nM), gebiotinyleerd substraatpeptide (30 nM), Fe(II) (1 μM), ascorbaat (100 μM), 2OG (10 μM) en werd uitgevoerd bij kamertemperatuur. Voor PHD2 bestond de reactie uit enzym (5 nM), gebiotinyleerd substraatpeptide (60nM), Fe(II) (20 μM), ascorbaat (200 μM), 2OG (2 μM) en werd uitgevoerd bij kamertemperatuur. EDTA wordt gebruikt om de reactie te stoppen (5 μL), AlphaScreen donor (Streptavidine-geconjugeerd) en acceptor (Proteïne A-geconjugeerd) kralen voorgeïncubeerd met peptideproductantilichamen worden toegevoegd (5 μL). Platen worden met folie afgedekt ter bescherming tegen licht, gedurende 60 minuten bij kamertemperatuur geïncubeerd en gelezen op een PHERAstar FS plaatlezer met behulp van een AlphaScreen 680 excitatie/570 emissiefilterset. De uiteindelijke kraalconcentratie in 20 μL reactie is 20 μg/mL. IC50-waarden worden berekend in Prism 6 na normalisatie tegen overeenkomstige DMSO-controles.

  • Methode

    Antiproliferative activities of compounds are determined by the MTT assay. HeLa cells are seeded into 96-well plates and cultured at 37 °C for 24 h to achieve 70%. Subsequently, the medium is replaced with DMEM medium containing the tested compounds in different concentrations of 1-300 μM in 1% DMSO. Staurosporine in 0.03-10 μM final concentration is used as a control for cytotoxicity. After 24 h of treatment, the medium is replaced with CellTiter 96 Aqueous One Solution Reagent and incubated for 4 hours. CC50 values are calculated in Prism 6 software after normalisation against corresponding 1% DMSO treated cells and 1% DMSO in media (no cells) controls.
