WZ4003 is een zeer specifieke NUAK kinase remmer met een IC50 van respectievelijk 20 nM en 100 nM voor NUAK1 en NUAK2 in celgebaseerde assays, zonder significante remming van 139 andere kinases.

In vitro

In HEK-293-cellen die wild-type NUAK1 tot expressie brengen, onderdrukt WZ4003 (3-10 μM) de NUAK1-gemedieerde MYPT1-fosforylering aanzienlijk. Bovendien remt deze verbinding (10 μM) de MYPT1 Ser445-fosforylering, evenals celmigratie, invasie en proliferatie in een vergelijkbare mate als knock-out in MEF's of knock-down in U2OS-cellen van NUAK1. Het vertoont ook een hoge, specifieke affiniteit voor de L858R/T790M-mutante EGFR, terwijl het een significant verminderde cellulaire IC50 vertoont tegen T790M-bevattende Ba/F3-cellen.

Protocol (uit referentie)

Kinase-assay:^{^[1]}	IC50-bepaling Actieve GST–NUAK1, GST–NUAK1[A195T] en GST–NUAK2 enzymen worden gezuiverd met behulp van glutathion–Sepharose uit HEK-293 cellysaten 36–48 uur na de transiënte transfectie van pEBG2T zoogdierconstructies die N-terminaal GST-getagde NUAK1, NUAK1[A195T] of NUAK2 tot expressie brengen. Voor peptidekinase-assays worden 96-well platen gebruikt en elke reactie wordt in drievoud uitgevoerd. Elke reactie wordt uitgevoerd in een totaal volume van 50 μL met 100 ng NUAK1 (wild-type of A195T-mutant) of NUAK2 in 50 mM Tris/HCl (pH 7,5), 0,1 mM EGTA, 10 mM magnesiumacetaat, 200 μM Sakamototide, 0,1 mM [γ -³²P]ATP (450–500 c.p.m./pmol) en de aangegeven concentraties remmers opgelost in DMSO. Na incubatie gedurende 30 min bij 30°C worden de reacties beëindigd door toevoeging van 25 mM (final) EDTA om het magnesium te cheleren. Vervolgens wordt 40 μL van het reactiemengsel op P81-papier gespot en ondergedompeld in 50 mM orthofosforzuur. De monsters worden driemaal gewassen in 50 mM orthofosforzuur, gevolgd door een enkele acetonspoeling en luchtdroging. De incorporatie van [γ -³²P]ATP in Sakamototide wordt gekwantificeerd door Cerenkov-telling. De waarden worden uitgedrukt als een percentage van de DMSO-controle. IC50-curves worden ontwikkeld en IC50-waarden worden berekend met GraphPad Prism software.
