세포 분석:

  • A549 및 H460 세포는 6-웰 플레이트(~106 세포/웰)에서 배양하고 두 그룹으로 나누었습니다. 모든 웰은 비특이적 결합을 차단하기 위해 5% BSA와 함께 배양되었습니다. 그룹 A는 124I-Durva-F(ab′)2의 결합을 차단하기 위해 100배 과량의 비표지 durvalumab으로 사전 포화되었고, 그룹 B는 배지만 추가되었습니다. 30분 후, 2 nM의 124I-Durva-F(ab′)2가 각 그룹에 첨가되어 세포 배양기에서 37 °C로 1시간 동안 배양되었습니다. 상층액 수집 후, 세포는 2 M NaOH를 사용하여 용해되었습니다.
