세포 분석:

  • TIL의 세포내 사이토카인 분석을 위해 해리된 종양 세포를 1×105~1×106 세포 밀도 범위로 96웰 U-바닥 플레이트에 펨브롤리주맙 10 µg/mL, 이필리무맙 10 µg/mL, 가용성 항-CD3 50 ng/mL, 항-CD28 50 ng/mL를 포함하여 37°C에서 24시간 동안 배양한 후 브레펠딘 A와 모넨신을 첨가했습니다. 항-CD3 및 항-CD28 자극 시 PD-1 발현에 따른 CD8+ TIL의 인터페론(IFN)-γ 및 종양 괴사 인자(TNF) 분비 수준을 측정하기 위해 브레펠딘 A와 모넨신 없이 24시간 동안 진행된 사전 배양 단계는 생략되었습니다. 12시간 후, 세포를 수확하고 염색했습니다. 세포내 사이토카인은 표면 염색, 고정 및 Foxp3 고정/투과 용액으로 투과 처리 후 염색했습니다.
