Torin1 inhibe la phosphorylation des substrats de mTORC1 et mTORC2 dans les cellules à des concentrations de 2 et 10 nM, respectivement. De plus, Torin1 présente une sélectivité 1000 fois supérieure pour mTOR par rapport à PI3K (EC50 = 1800 nM) et une sélectivité de liaison 100 fois supérieure par rapport à 450 autres protéines kinases. Torin1 provoque un arrêt du cycle cellulaire via un mécanisme résistant à la rapamycine qui est également indépendant de mTORC2. Torin1 perturbe les phénotypes dépendants de mTORC1 plus complètement que la rapamycine. Les fonctions de mTORC1 résistantes à la rapamycine sont nécessaires pour la traduction dépendante de la coiffe. Dans une étude récente, il est rapporté que Torin1 augmente la sécrétion et l'expression génique de la neurotensine par l'activation de la voie MEK/ERK/c-Jun dans la lignée cellulaire endocrine humaine BON.

Pour produire du mTORC1 soluble, des lignées cellulaires HEK-293T exprimant de manière stable le Raptor marqué FLAG en N-terminal sont générées à l'aide d'un rétrovirus MSCV pseudotypé par le virus de la stomatite vésiculeuse G. Pour mTORC2, des cellules HeLa similaires exprimant de manière stable le Protor-1 marqué FLAG en N-terminal sont générées. Les deux complexes sont purifiés en lysant les cellules dans 50 mm HEPES, pH 7,4, 10 mm pyrophosphate de sodium, 10 mm
micro-glycérophosphate de sodium, 100 mm NaCl, 2 mm EDTA, 0,3% CHAPS. Les cellules sont lysées à 4 °C pendant 30 min, et la fraction insoluble est retirée par microcentrifugation à 13 000 tr/min pendant 10 min. Les surnageants sont incubés avec de l'anticorps monoclonal FLAG-M2-agarose pendant 1 h, puis lavés trois fois avec du tampon de lyse et une fois avec du tampon de lyse contenant une concentration finale de 0,5 M NaCl. Le mTORC1 purifié est élué avec 100
microg/mL de peptide 3
cFLAG dans 50 mm HEPES, pH 7,4, 100 mm NaCl. L'éluat peut être aliquote et stocké à -80 °C. Les substrats S6K1 et Akt1 sont purifiés. Les tests de kinase sont réalisés pendant 20 min à 30 °C dans un volume final de 20
microL composé du tampon de kinase (25 mm HEPES, pH 7,4, 50 mm KCl, 10 mm MgCl
2, 500
microM ATP) et de 150 ng de S6K1 ou Akt1 inactif comme substrats. Les réactions sont arrêtées par l'ajout de 80
microL de tampon d'échantillon et bouillies pendant 5 min. Les échantillons sont ensuite analysés par SDS-PAGE et immunobuvardage.

Torin1 est efficace à une dose de 20 mg/kg dans un modèle de xénogreffe U87MG et démontre une bonne inhibition pharmacodynamique des effecteurs en aval de mTOR dans les tissus tumoraux et périphériques.
